Project Title: An investigation of the aggregation propensity of some model proteins and their modulation by cosolvents. Shashank Deep Assosiate Professor (Physical chemistry/ Biophysical Chemistry)

The main objective of this proposal is to advance the understanding of physico-chemical basis of the protein-protein self association and mechanism of aggregation. To this end, we plan a systematic structural characterization and thermodynamic investigations of model proteins under the condition of aggregation using various experimental and computational tools. Structural and thermodynamic/energetics aspects are fundamental and provide the indispensable background for the understanding of the functional and dynamic properties of the system. We also plan to study the kinetics of aggregation since they give indispensable information to decipher the mechanism of aggregation. The concentration, temperature and pressure dependence of various kinetics parameters will be probed to obtain the minute details of the aggregation processes. To effectively inhibit aggregation, both in vivo and in vitro, a more complete understanding of the mechanisms by which proteins aggregate and by which varying solution conditions affect this process is needed. Thus we plan to carry out these investigations in presence of sugars, polyols and amine additives. The change of different structural/kinetic/thermodynamic parameters associated with the protein aggregation in presence of cosolvents obtained trough experimental and computational approaches will be correlated to role of nonnative conformations; stability of these conformations, physico-chemical properties of solvent like surface tension and viscosity that may lay the groundwork for future therapeutic approach. ,

Brief plan of research
The research activities of our group include “chiron approach” towards the synthesis of biologically and pharmacologically important compounds from

commercially available and cheap carbohydrates as starting materials. Recently, our group has developed a simple route towards the synthesis of N-linked glycoamino acids and glycopeptides, compounds that play crucial role in cell-cell communications, cell adhesion, signal transduction etc. Total syntheses of natural products such as pochonicine, balanol, steviamine, conduramines etc. and their novel synthetic analogues through “diversity oriented synthesis” are currently being pursued in our lab. Other interests include the development of carbohydrate based asymmetric catalysts as well as synthesis and use of chiral N-heterocyclic carbenes as catalysts for enantioselective organic reactions. The student will have the opportunity to learn all the techniques of organic synthesis as well as their characterization.

Organic Synthesis
Nidhi Jain The research is focused towards the development of new reaction methodologies in synthetic organic chemistry, in particular transition-metal catalyzed reactions involving carbon-carbon and carbon-hetero atom bond formation for the synthesis of medicinally relevant heterocycles. A major interest is to activate a specific 'inert' C-H bond and transform it to a more versatile functional group under ambient conditions to form a diverse range of useful molecular architectures. The biggest challengehere is to identify novel strategies for the formation of organo-metal intermediates under mild conditions, and to discover new methods for their subsequent functionalisation.
