Hypothesis

1) If a calculated absorbance level is directly related to the concentration of albumin, then a higher concentration of albumin will lead to higher absorbance level.

Results

Table No.| Albumin(cm3)| KCl (mL)| Biuret Reagent (mL)| Absorbance| 1| 0| 5| 3| 0.049|
2| 1| 4| 3| 0.112|
3| 2| 3| 3| 0.258|
4| 4| 1| 3| 0.39|
5| 5| 0| 3| 0.418|
6| 5 (A)| 0| 3| 0.198|
7| 5 (B)| 0| 3| 0.347|
Table 1) Test Result

Table 1 is the results of seven test tubes with a different amount of solutions. 3 mL of Biuret Reagent was added to all seven test tubes and absorbance was measured with spectrophotometer. It is observed that the absorbance level increases as the amount of albumin is added more in different test tubes, while the amount of KCl decreases.

Concentration of albumin in mg(x-axis)| Absorbance (y-axis)| 0| 0.049|
2.5| 0.112|
5| 0.258|
10| 0.39|
12.5| 0.418|
(A)| 0.198|
(B)| 0.347|
Table 2) Analyze of two factors, concentration of albumin and absorbance

The concentration of albumin in the standard albumin is 2.5 mg/cm3of 0.5 M KCl. Hence 1.0 cm3of albumin is calculated to 2.5 mg of the concentration of albumin. For 6th and 7th test tube were unknown labeled as (A) and (B).
